making 6s out of 4s+2s?

I just purchased a esky E-smart, and am planning on running it with 6s. I have 2 brand new 4s 4500mah batteries, and just bought two 2s 4500mah batteries (same brand as the 4s) to make up 2 complete 6s 4500mah batteries. This will be ok, right? And as far as charging, can I charge both the 4s & 2s together, charging each through their own balancer, as a 6s pack, or do I need to charge them separately?

Using a series harness is quite common. I made one of my own to use a 2s and 3s pack for 5s. See photo below...

Charging... I'll charge mine separately as individual packs purely as a safety precaution.
